Types of Stem Cells

Stem cells are categorized in 2 ways:

Origin-based classification describes where the stem cells come from.

Potency or differentiation potential-based describes what the stem cells can become.

Scientists use both because each system highlights different important aspects of stem cells.

Differentiation potential-based classification

Classification by differentiation refers to the types of other cells that stem cells can mature into (ordered from broad to specific):

  • Totipotent develops into any cell type in the body and extra-embryonic tissues like the placenta (example: the zygote and very early embryonic cells)
  • Pluripotent becomes almost any cell type in the body but not extra-embryonic tissues (embryonic stem cells and induced pluripotent stem cells fall into this category)
  • Multipotent differentiates into several related cell types within a group (example: blood stem cells can become red blood cells, white blood cells, or platelets, but they can’t become brain cells or skin cells)
  • Oligopotent differentiates into fewer, closely related cell types (a narrower subset of multipotent)
  • Unipotent can produce only 1 cell type but retain the ability to self-renew (example: muscle stem cells)

These all end in ‘-potent,’ signifying the power each has to turn into other cell types.

Origin-based classification

Origin-based classification considers three types of stem cells:

  • Adult stem cells
  • Embryonic stem cells
  • Induced pluripotent stem cells

Adult stem cells help maintain and repair tissues throughout life.

They’re found in various tissues like bone marrow, brain, skin, and liver (i.e., liver stem cells will only produce liver cells).

They are usually multipotent or unipotent, meaning they generate only certain types of cells related to the tissue they originate from.

Embryonic stem cells come from early-stage embryos (unused embryos from in vitro fertilization donated for research). They’re pluripotent, developing into almost any cell type in the body.

Induced pluripotent stem cells (iPSCs) are adult cells genetically reprogrammed in the lab to behave like embryonic stem cells. They have the potential to become many cell types but their full capabilities are still under study.

Where are stem cells located?

Stem cells are found throughout the human body but tend to concentrate in particular tissues or “niches.”

They remain mostly dormant until needed for tissue maintenance or repair.

Key locations include:

  • Bone marrow: Contains hematopoietic stem cells (HSCs) that produce blood and immune cells, as well as mesenchymal stem cells (MSCs) that can become bone, cartilage, and fat cells
  • Umbilical cord blood and tissue: Rich in stem cells collected at birth, useful for regenerative medicine
  • Dental pulp: Inside teeth, containing MSCs
  • Fat (adipose) tissue: Contains MSCs that can be harvested for therapies

Finally, don’t forget proper hydration. Water is essential for every cellular process, including stem cell activity.

A good rule of thumb is to drink half your body weight in ounces daily. For example, I am 200 pounds, so I should aim to consume at least 100 ounces of water every day.

Fasting

Fasting is perhaps the most powerful stem cell regeneration tool available.

A 2020 research found that fasting increases the percentage of stem cells with long telomeres (Aging). Telomeres are “protective caps” at the end of chromosomes, preventing them from deteriorating.

This same study further found that fasting also increases the maximum telomere length. In flatworm (planarian) stem cells, anyway. But this opens up more understanding of stem cell aging and regeneration.

Meanwhile, MIT researchers discovered that mice fasting for 24 hours dramatically improved intestinal stem cells’ ability to regenerate (Cell Stem Cell).

During fasting, intestinal stem cells (ISCs) switch from burning glucose to burning fat, which triggers a metabolic shift that enhances their regenerative capacity (Cell Stem Cell).

However, there’s a downside. Regeneration also increased the risk of developing intestinal tumors when cancerous mutations happened during the regenerative period (Nature).

Columbia Stem Cell Initiative researchers found that the combination of fasting followed by refeeding rejuvenates old blood stem cells (HSCs) (Cell Stem Cell).

This makes them nearly as effective as young stem cells at creating new blood cells.

This 24-hour fasting-refeeding cycle activates your body’s cellular cleanup process (autophagy). Autophagy removes damaged components and resets stem cell function (Nature).

Exercise

Exercise promotes tissue regeneration primarily through stem cell activation and proliferation (J Tissue Eng Regen Med).

When you move your body, you activate and multiply mesenchymal stem cells (MSCs). These are the stem cells that differentiate into bone, cartilage, fat, and muscle cells.

University of Illinois researchers found that MSCs proliferate during exercise, contributing to stronger bones and muscle structures (MSSE J).
